Rhamnose-based glycomimetic for recruitment of endogenous anti-rhamnose antibodies

چکیده
Recruitment of natural antibodies towards tumour cells for their elimination by the immune system could be a highly specific and efficacious therapeutic strategy. While l-rhamnose has already been explored as suitable antigen antibody recruitment, we here report first rhamnose-based glycomimetic to used such purpose. The is designed more hydrolytically enzymatically stable than rhamnosides, provides site easy further conjugation proved capture anti-rhamnose IgG in serological ELISA assay.

Rhamnose glycoconjugates for the recruitment of endogenous anti-carbohydrate antibodies to tumor cells.
Immunotherapy is a promising strategy for targeting tumors. One emerging approach is to harness the immune effector functions of natural antibodies to destroy tumor cells. Dinitrophenyl (DNP) and the galactose-α-1,3-galactose (αGal) epitope are two haptens that bind endogenous antibodies. One potential alternative is the deoxysugar L-rhamnose. We compared these candidates by using a biosensor a...

The rhamnose utilization pathway in Pichia pastoris has not been clarified although this strain can grow well on rhamnose as a sole carbon source. In this study, four genes, PAS_chr4_0338, PAS_chr4_0339, PAS_chr4_0340, and PAS_chr4_0341, were, for the first time, predicted to be involved in rhamnose metabolism along with the previously identified gene PAS_chr1_4-0075. Moreover, expression of th...
متن کاملMetabolism of L-rhamnose by Escherichia coli. I. L-rhamnose isomerase.
The methyl pentose, L-rhamnose, is known to be fermented by a number of bacteria, but little information is available concerning the metabolic steps involved in this fermentation. An indication of the pathway of rhamnose metabolism was provided by the finding of Kluyver and Schnellen (1937) that propylene glycol was a major product in the fermentation by Bacillus rhamnosifermentans. Since 50 pe...
متن کاملFlavonol rhamnosylation indirectly modifies the cell wall defects of RHAMNOSE BIOSYNTHESIS1 mutants by altering rhamnose flux.
Rhamnose is required in Arabidopsis thaliana for synthesizing pectic polysaccharides and glycosylating flavonols. RHAMNOSE BIOSYNTHESIS1 (RHM1) encodes a UDP-l-rhamnose synthase, and rhm1 mutants exhibit many developmental defects, including short root hairs, hyponastic cotyledons, and left-handed helically twisted petals and roots. It has been proposed that the hyponastic cotyledons observed i...
